These results reveal a novel role for Cx43 in coordinating cell dedifferentiation and redifferentiation in osteoarthritis to manage extracellular matrix remodelling and to restrain senescence and inflammation. Among the cascade of events leading to osteoarthritic chondrocytes reprogramming, upregulation of Cx43 increases the degree of cellular dedifferentiation towards an immature state via chondrocyte-mesenchymal transition. Thus, drugs targeting Cx43 to induce redifferentiation may protect against cartilage degradation in osteoarthritis and boost regeneration.
